PRINCIPLE OF THE ASSAY Human MSP/MST1 ELISA Kit is based on the quantitative sandwich enzyme-linked immunosorbent assay technique to measure concentration of human MSP in the samples. An antibody specific for human MSP has been immobilized onto microwells. Standard or samples are pipetted into the wells, followed by the addition of biotin-linked detect antibody specific for MSP, and MSP present is bound by the immobilized antibody and detect antibody following the first incubation. After removal of any unbound substances, streptavidin-HRP is added for a second incubation. After washing, substrate solution reacts with HRP and color develops in proportion to the amount of MSP bound by the immobilized antibody. The color development is stopped by addition of acid and the optical density value is measured by microplate reader.
Describtion

MSP (Macrophage Stimulating Protein; also known as Hepatocyte Growth Factor-like protein (HGFL) and MST1) is a member of the HGF family of growth factors .MSP is secreted as an inactive single chain precursor (pro-MSP) ,and is a secreted protein and the ligand for transmembrane receptor tyrosine kinase Recepteur d'Origine Nantais (RON; also known as MST1R). Since its discovery, MSP has been demonstrated to play a key role in regulating inflammation in the peripheral tissues of multiple disease models. Recent evidences also point toward a beneficial role of MSP in the regulation of hepatic lipid and glucose metabolism, thereby implicating MSP as a crucial regulator in maintaining metabolic homeostasis while simultaneously suppressing inflammatory processes. MSP induces macrophage and keratinocyte proliferation and osteoclast activation. It also inhibits the expression of iNOS and IL-12 induced by LPS or IFN through macrophages, and can prevent apoptosis of epithelial cells isolated from the extracellular matrix.
