PRINCIPLE OF THE ASSAY | Mouse gp130 ELISA Kit is based on the quantitative sandwich enzyme-linked immunosorbent assay technique to measure concentration of mouse gp130 in the samples. An antibody specific for mouse gp130 has been immobilized onto microwells. Standard or samples are pipetted into the wells, followed by the addition of biotin-linked detect antibody specific for gp130, and gp130 present is bound by the immobilized antibody and detect antibody following the first incubation. After removal of any unbound substances, streptavidin-HRP is added for a second incubation. After washing, substrate solution reacts with HRP and color develops in proportion to the amount of gp130 bound by the immobilized antibody. The color development is stopped by addition of acid and the optical density value is measured by microplate reader. |
Describtion | Glycoprotein 130 (also known as gp130, IL6ST, IL6-beta or CD130), the common signal transducing receptor component shared by the functional receptor complexes of the IL-6 family of cytokines, belongs to the class I cytokine receptor family. The protein is a signal transducer shared by many cytokines, including interleukin 6 (IL6), ciliary neurotrophic factor (CNTF), leukemia inhibitory factor (LIF), and oncostatin M (OSM). The activation of this protein is dependent upon the binding of cytokines to their receptors. Binding of IL-6 or IL-11 to either the membrane-anchored or soluble IL-6 R or IL-11 R, respectively, initiates the association of IL-6 R (IL-11 R) with gp130. For other IL-6 family cytokines, such as LIF and OSM, signal transduction is similarly triggered by the heterodimerization of gp130 with LIFR or OSM R. Knockout studies in mice suggested a critical role of the gene encoding this protein in regulating myocyte apoptosis. |
