PRINCIPLE OF THE ASSAY | Mouse Prolactin ELISA Kit is based on the quantitative sandwich enzyme-linked immunosorbent assay technique to measure concentration of mouse Prolactin in the samples. An antibody specific for mouse Prolactin has been immobilized onto microwells. Standard or samples are pipetted into the wells, followed by the addition of biotin-linked detect antibody specific for Prolactin, and Prolactin present is bound by the immobilized antibody and detect antibody following the first incubation. After removal of any unbound substances, streptavidin-HRP is added for a second incubation. After washing, substrate solution reacts with HRP and color develops in proportion to the amount of Prolactin bound by the immobilized antibody. The color development is stopped by addition of acid and the optical density value is measured by microplate reader. |
Describtion | Prolactin (PRL) is a neuroendocrine pituitary hormone, encoded by the PRL gene,that is synthesized by anterior pituitary, placenta, brain, uterus, dermal fibroblasts, decidua, B cells, T cells, natural killer cells, and breast cancer cells. Prolactin expression is low during early human pregnancy, replaced by placental lactogen, but increases in late pregnancy. Prolactin plays an essential role in metabolism, regulation of the immune system and pancreatic development. Prolactin also acts in a cytokine-like manner and as an important regulator of the immune system. It has important cell cycle-related functions as a growth-, differentiating- and anti-apoptotic factor. As a growth factor, binding to cytokine-like receptors, it influences hematopoiesis, angiogenesis and is involved in the regulation of blood clotting through several pathways. |
