PRINCIPLE OF THE ASSAY | Rat Adiponectin/Acrp30 ELISA Kit is based on the quantitative sandwich enzyme-linked immunosorbent assay technique to measure concentration of rat Adiponectin in the samples. An antibody specific for rat Adiponectin has been immobilized onto microwells. Standard or samples are pipetted into the wells, followed by the addition of biotin-linked detect antibody specific for Adiponectin, and Adiponectin present is bound by the immobilized antibody and detect antibody following the first incubation. After removal of any unbound substances, streptavidin-HRP is added for a second incubation. After washing, substrate solution reacts with HRP and color develops in proportion to the amount of Adiponectin bound by the immobilized antibody. The color development is stopped by addition of acid and the optical density value is measured by microplate reader. |

Adiponectin, also known as Adipocyte complement- related protein of 30 kDa (Acrp30), is a protein involved in regulating glucose levels as well as fatty acid breakdown. Adiponectin assembles into different complexes including trimers, hexamers, and higher order oligomeric structures that may affect biological activity. Adiponectin is a protein hormone that modulates a number of metabolic processes, including glucose regulation and fatty acid oxidation and is exclusively secreted from adipose tissue (and also from the placenta in pregnancy) into the bloodstream and is very abundant in plasma relative to many hormones. Adiponectin may also play anti-atherogenic and anti- inflammatory roles, and negatively regulates myelomonocytic progenitor cell growth and TNF-α production in macrophages. Monitoring of Adiponectin levels in serum provides more detailed insights in several pathological situations such as type 2 diabetes, obesity, atherosclerosis and non-alcoholic fatty liver disease. |
