Whole Blood Sample: Take 100 μl of anticoagulant and add 5 ul antibodies. Shake well and incubate at room temperature in the dark for 15 minutes or at 2-8℃ in the dark for 30 minutes. Follow the instructions of the red blood cell lysis reagent to remove red blood cells. Centrifuge at 300 xg for 5-10 minutes, discard the supernatant, resuspend in 500 μl of flow cytometry staining buffer, and perform machine detection.
For other samples: Take 1×10^5 – 10^8 single cells after appropriate centrifugation, resuspend in an appropriate amount of flow cytometry staining buffer (Catalog No. S1001), add 5 ul antibodies to make a final reaction volume of 100 μl. Shake well and incubate at room temperature in the dark for 15 minutes or at 2-8℃ in the dark for 30 minutes. Add 1-2 ml of flow cytometry staining buffer to each tube, centrifuge at 300 × g for 5-10 minutes, discard the supernatant, resuspend in 500 μl of flow cytometry staining buffer, and perform machine detection.
