Assay Principle | This kit utilizes the double antibody sandwich enzyme-linked immunosorbent assay (ELISA) detection technique.Specific anti-mouse IL-21 antibodies are precoated on a high-affinity ELISA plate. Standard samples, test samples, and biotinylated detection antibodies are added to the wells of the ELISA plate. After incubation, IL-21 present in the samples binds to the solid-phase antibodies and the detection antibodies. After washing to remove unbound substances, streptavidin-HRP labeled with horseradish peroxidase is added. After washing, a colorimetric substrate, TMB, is added and the plate is incubated in the dark for color development. The intensity of the color reaction is directly proportional to the concentration of IL-21 in the samples.A stop solution is added to terminate the reaction, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm). |

Human IL21 Predicted Functions
Enables interleukin-2 receptor binding activity. Involved in several processes, including positive regulation of interleukin-17 production; positive regulation of natural killer cell mediated cytotoxicity; and tyrosine phosphorylation of STAT protein. Acts upstream of or within positive regulation of B cell proliferation; positive regulation of T cell proliferation; and positive regulation of tyrosine phosphorylation of STAT protein. Predicted to be located in extracellular region. Implicated in asthma; common variable immunodeficiency; hepatocellular carcinoma; and systemic lupus erythematosus. Biomarker of several diseases, including anogenital venereal wart; autoimmune disease (multiple); dengue hemorrhagic fever; human immunodeficiency virus infectious disease; and tropical spastic paraparesis.

Mouse Il21 Predicted Functions
Enables cytokine receptor binding activity. Involved in cellular response to virus; lymphocyte activation involved in immune response; and positive regulation of immunoglobulin production. Acts upstream of or within positive regulation of cytokine production; positive regulation of natural killer cell differentiation; and positive regulation of natural killer cell mediated cytotoxicity. Predicted to be located in extracellular space. Is expressed in ductus deferens; epididymis; and testis. Human ortholog(s) of this gene implicated in asthma; common variable immunodeficiency; hepatocellular carcinoma; and systemic lupus erythematosus. Orthologous to human IL21 (interleukin 21).
