Assay PrincipleThis kit utilizes the double antibody sandwich enzyme-linked immunosorbent assay (ELISA) detection technique.Specific anti-human MPO antibodies are precoated on a high-affinity ELISA plate. Standard samples, test samples, and biotinylated detection antibodies are added to the wells of the ELISA plate. After incubation, MPO present in the samples binds to the solid-phase antibodies and the detection antibodies. After washing to remove unbound substances, streptavidin-HRP labeled with horseradish peroxidase is added. After washing, a colorimetric substrate, TMB, is added and the plate is incubated in the dark for color development. The intensity of the color reaction is directly proportional to the concentration of MPO in the samples.A stop solution is added to terminate the reaction, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm).

Human MPO Target Information

Human MPO Predicted Functions

Enables heparin binding activity and peroxidase activity. Involved in low-density lipoprotein particle remodeling. Acts upstream of or within hydrogen peroxide catabolic process. Located in azurophil granule; extracellular space; and nucleoplasm. Implicated in several diseases, including Alzheimer’s disease 1; cystic fibrosis; kidney failure (multiple); liver disease (multiple); and lung disease (multiple). Biomarker of asthma (multiple); glomerulonephritis; and lung disease (multiple).

Mouse Mpo Target Information

Mouse Mpo Predicted Functions

Enables peroxidase activity. Acts upstream of or within several processes, including hypochlorous acid biosynthetic process; removal of superoxide radicals; and response to fungus. Located in mitochondrion. Is expressed in several structures, including alimentary system; genitourinary system; hemolymphoid system; integumental system; and skeleton. Human ortholog(s) of this gene implicated in several diseases, including Alzheimer’s disease 1; cystic fibrosis; kidney failure (multiple); liver disease (multiple); and lung disease (multiple). Orthologous to human MPO (myeloperoxidase).

Rat Mpo Target Information

Rat Mpo Predicted Functions

Enables peroxidase activity. Involved in several processes, including response to food; response to gold nanoparticle; and response to lipopolysaccharide. Located in extracellular space. Used to study acute kidney failure. Biomarker of chronic obstructive pulmonary disease; cystitis; and renovascular hypertension. Human ortholog(s) of this gene implicated in several diseases, including Alzheimer’s disease 1; cystic fibrosis; kidney failure (multiple); liver disease (multiple); and lung disease (multiple). Orthologous to human MPO (myeloperoxidase).
