Assay PrincipleThis kit utilizes the double antibody sandwich enzyme-linked immunosorbent assay (ELISA) detection technique.Specific anti-human TIMP-3 antibodies are precoated on a high-affinity ELISA plate. Standard samples, test samples, and biotinylated detection antibodies are added to the wells of the ELISA plate. After incubation, TIMP-3 present in the samples binds to the solid-phase antibodies and the detection antibodies. After washing to remove unbound substances, streptavidin-HRP labeled with horseradish peroxidase is added. After washing, a colorimetric substrate, TMB, is added and the plate is incubated in the dark for color development. The intensity of the color reaction is directly proportional to the concentration of TIMP-3 in the samples.A stop solution is added to terminate the reaction, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm).

Human TIMP3 Predicted Functions

Predicted to enable metalloendopeptidase inhibitor activity and protease binding activity. Involved in negative regulation of ERK1 and ERK2 cascade; negative regulation of membrane protein ectodomain proteolysis; and positive regulation of TRAIL-activated apoptotic signaling pathway. Located in nucleus. Colocalizes with collagen-containing extracellular matrix. Implicated in Sorsby’s fundus dystrophy; breast carcinoma; and urinary bladder cancer. Biomarker of several diseases, including breast cancer (multiple); preretinal fibrosis; reproductive organ cancer (multiple); thoracic aortic aneurysm; and type 2 diabetes mellitus.

Mouse Timp3 Predicted Functions

Predicted to enable metalloendopeptidase inhibitor activity and protease binding activity. Involved in negative regulation of metalloendopeptidase activity. Acts upstream of or within cellular response to organic substance. Located in basement membrane. Is expressed in several structures, including alimentary system; central nervous system; eye posterior segment; genitourinary system; and musculoskeletal system. Used to study Sorsby’s fundus dystrophy and dilated cardiomyopathy. Human ortholog(s) of this gene implicated in Sorsby’s fundus dystrophy; breast carcinoma; and urinary bladder cancer. Orthologous to human TIMP3 (TIMP metallopeptidase inhibitor 3).
