THBS1

HGNC ID: 11785

Target Abstract:

Thrombs-1 (THBS-1) is a member of the thrombs-retin family that mediates cell-cell and cell-matrix interactions. The protein can bind to fibrinogen, fipronectin, laminin, type V collagen and integrin α-V/β-1. It plays a role in platelet aggregation, angiogenesis and tumorigenesis. In normal tissues, it is a natural inhibitor of new blood vessel formation and tumorigenesis. THBS-1 also positively and negatively regulates endothelial cell adhesion, movement and growth. By blocking the binding of THBS-1 to the cell surface receptor (CD47), normal tissues become almost immune to tumor radiation therapy and contribute to tumor death.

Human THBS1 Predicted Functions

Enables several functions, including collagen V binding activity; fibrinogen binding activity; and fibroblast growth factor binding activity. Involved in several processes, including regulation of cell migration; regulation of gene expression; and regulation of signal transduction. Acts upstream of or within several processes, including negative regulation of endothelial cell chemotaxis; negative regulation of endothelial cell proliferation; and negative regulation of nitric oxide mediated signal transduction. Located in several cellular components, including external side of plasma membrane; extracellular space; and platelet alpha granule. Part of fibrinogen complex. Colocalizes with collagen-containing extracellular matrix. Implicated in cholangiocarcinoma; pancreatic cancer; and pancreatic ductal carcinoma. Biomarker of COVID-19; cholangiocarcinoma; obesity; pancreatic cancer (multiple); and urinary bladder cancer.

Mouse Thbs1 Target Information

Mouse Thbs1 Predicted Functions

Enables extracellular matrix binding activity. Involved in several processes, including behavioral response to pain; positive regulation of macrophage chemotaxis; and positive regulation of transforming growth factor beta receptor signaling pathway. Acts upstream of or within several processes, including cellular response to nitric oxide; circulatory system development; and monocyte aggregation. Located in extracellular space and sarcoplasmic reticulum. Is expressed in several structures, including alimentary system; brain; genitourinary system; limb; and sensory organ. Used to study Sjogren’s syndrome. Human ortholog(s) of this gene implicated in cholangiocarcinoma; pancreatic cancer; and pancreatic ductal carcinoma. Orthologous to human THBS1 (thrombospondin 1).

Rat Thbs1 Target Information

Rat Thbs1 Predicted Functions

Enables transforming growth factor beta binding activity. Involved in several processes, including cellular response to tumor necrosis factor; positive regulation of transforming growth factor beta production; and response to testosterone. Located in cytoplasm and extracellular space. Biomarker of diabetic retinopathy; heart disease; myocardial infarction; and prostate cancer. Human ortholog(s) of this gene implicated in cholangiocarcinoma; pancreatic cancer; and pancreatic ductal carcinoma. Orthologous to human THBS1 (thrombospondin 1).
