Target Abstract:
Interleukin 33 (IL-33), also known as NF-HEV and DVS27, is a member of the IL-1 family with a molecular weight of 30 kDa, and is secreted by a variety of cell types including fibroblasts, mast cells, dendritic cells, macrophages, osteoblasts, endothelial cells, and epithelial cells.IL-33 induces the secretion of type II cytokines by helper T cells, mast cells, eosinophils, and basophils. Type II cytokines are secreted by granulocytes.IL-33 mediates its biological functions by binding to the receptor ST2 and IL-1 receptor accessory protein (IL1RAP) and activating extracellular molecules in the NF-κB and MAP kinase signaling pathways that can induce the secretion of type II cytokines (e.g., IL-5 and IL-13) from polarized Th2 cells. After injection of IL-33, it induces type II cytokines in vivo, which is thought to induce severe pathological changes in mucosal organs.

Human IL33 Predicted Functions
Enables cytokine activity. Involved in negative regulation of transcription by RNA polymerase II; positive regulation of chemokine production; and positive regulation of macrophage activation. Located in nucleoplasm. Part of cytoplasm and nucleus. Implicated in candidiasis; inflammatory bowel disease; and peptic ulcer disease. Biomarker of several diseases, including colitis (multiple); common cold; gout; liver disease (multiple); and tuberculosis (multiple).

Mouse Il33 Predicted Functions
Enables cytokine activity and interleukin-33 receptor binding activity. Involved in several processes, including interleukin-33-mediated signaling pathway; microglial cell activation involved in immune response; and regulation of gene expression. Acts upstream of or within several processes, including negative regulation of macrophage proliferation; positive regulation of cellular defense response; and positive regulation of macromolecule metabolic process. Located in nucleus. Is expressed in several structures, including adrenal gland; alimentary system; genitourinary system; nervous system; and sensory organ. Used to study Alzheimer’s disease. Human ortholog(s) of this gene implicated in candidiasis; inflammatory bowel disease; and peptic ulcer disease. Orthologous to human IL33 (interleukin 33).
