Assay PrincipleThis kit utilizes the double antibody sandwich enzyme-linked immunosorbent assay (ELISA) detection technique.Specific anti-mouse IL-33 antibodies are precoated on a high-affinity ELISA plate.Standards and test samples are added to the wells of the ELISA plate. After incubation, the IL-33 present in the samples binds to the solid-phase antibodies. After washing to remove unbound substances, biotinylated detection antibodies are added and incubated. After washing to remove unbound biotinylated antibodies, streptavidin-HRP labeled with horseradish peroxidase is added. After washing again, a signal enhancer is added and incubated. After washing to remove unbound substances, Streptavidin-HRP is added once more. After washing, a colorimetric substrate, TMB, is added and the plate is incubated in the dark for color development. The intensity of the color reaction is directly proportional to the concentration of IL-33 in the samples. A stop solution is added to terminate the reaction, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm).

Human IL33 Predicted Functions

Enables cytokine activity. Involved in negative regulation of transcription by RNA polymerase II; positive regulation of chemokine production; and positive regulation of macrophage activation. Located in nucleoplasm. Part of cytoplasm and nucleus. Implicated in candidiasis; inflammatory bowel disease; and peptic ulcer disease. Biomarker of several diseases, including colitis (multiple); common cold; gout; liver disease (multiple); and tuberculosis (multiple).

Mouse Il33 Target Information

Mouse Il33 Predicted Functions

Enables cytokine activity and interleukin-33 receptor binding activity. Involved in several processes, including interleukin-33-mediated signaling pathway; microglial cell activation involved in immune response; and regulation of gene expression. Acts upstream of or within several processes, including negative regulation of macrophage proliferation; positive regulation of cellular defense response; and positive regulation of macromolecule metabolic process. Located in nucleus. Is expressed in several structures, including adrenal gland; alimentary system; genitourinary system; nervous system; and sensory organ. Used to study Alzheimer’s disease. Human ortholog(s) of this gene implicated in candidiasis; inflammatory bowel disease; and peptic ulcer disease. Orthologous to human IL33 (interleukin 33).

Rat Il33 Target Information

Rat Il33 Predicted Functions

Predicted to enable cytokine activity and interleukin-33 receptor binding activity. Involved in several processes, including cellular response to mechanical stimulus; positive regulation of oligodendrocyte differentiation; and regulation of sensory perception of pain. Located in nucleus. Biomarker of acute myocardial infarction; colon cancer; and diabetes mellitus. Human ortholog(s) of this gene implicated in candidiasis; inflammatory bowel disease; and peptic ulcer disease. Orthologous to human IL33 (interleukin 33).
