Target Abstract: Carboxypeptidase B1 (CPB1), also known as Procarboxypeptidase B (PCPB), is a pancreas-specific protein. It has been validated as a serum biomarker for acute pancreatitis and pancreatic transplant rejection. As a representative exopeptidase that cleaves C-terminal basic residues of proteins, CPB1 specifically acts on C-terminal arginine and lysine residues, with a preference for arginine. The B1 and B2 isoforms of procarboxypeptidase B differ primarily in isoelectric point. Predicted from its amino acid sequence, the precursor protein of procarboxypeptidase B consists of a 15-amino-acid signal peptide, a 95-amino-acid activation peptide, and a 37-amino-acid mature protein chain, totaling 416 amino acids. The secreted procarboxypeptidase B precursor requires limited proteolysis by trypsin to be converted into enzymatically active CPB1.
