Target Abstract:
ZAP-7, also known as TCR-associated protein-70, is expressed in T cells and NK cells and is required for TCR signaling and development.ZAP-70 interacts with the TCR complex by binding to ITAMs present in the invariant subunit of the TCR complex. Upon activation, ZAP-70 is phosphorylated on several tyrosine residues by two mechanisms: autophosphorylation and trans-phosphorylation by the Src family tyrosine kinase Lck1-3. Tyrosine phosphorylation of ZAP-70 is associated with its increased kinase activity and triggers downstream signaling events. Although ZAP-70 is normally expressed in T cells and NK cells, several recent studies have also shown that positive ZAP-70 expression is highly correlated with mutant IgVH expression in B chronic lymphocytic leukemia (CCL). In conclusion, ZAP-70 expression can be measured by intracellular flow cytometry and can be used as a prognostic indicator in B-CCL.
