Assay PrincipleThis kit utilizes a double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) technique. Specific anti-human ErbB3 antibodies are pre-coated on a high-affinity ELISA plate. Standard samples, test samples, and detection antibodies labeled with horseradish peroxidase (HRP) are added to the wells of the ELISA plate. After incubation, the ErbB3 present in the sample binds to the solid-phase antibody and the detection antibody. After washing, the TMB chromogenic substrate is added for color development while avoiding light. The intensity of the color reaction is directly proportional to the concentration of ErbB3 in the sample. The reaction is terminated by adding a stop solution, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm).

Human Predicted Functions

Enables several functions, including ErbB-3 class receptor binding activity; neuregulin binding activity; and protein heterodimerization activity. Involved in several processes, including extrinsic apoptotic signaling pathway in absence of ligand; phosphatidylinositol 3-kinase signaling; and positive regulation of protein tyrosine kinase activity. Located in basolateral plasma membrane and extracellular space. Part of ERBB3:ERBB2 complex. Implicated in lethal congenital contracture syndrome 2; lung adenocarcinoma; lung non-small cell carcinoma; and neuronal intestinal dysplasia type A. Biomarker of carcinoma (multiple); ductal carcinoma in situ; prostate cancer; and urinary bladder cancer.

Enables neuregulin receptor activity. Involved in ERBB2-ERBB3 signaling pathway; Schwann cell development; and myelination. Acts upstream of or within several processes, including negative regulation of motor neuron apoptotic process; nervous system development; and positive regulation of calcineurin-NFAT signaling cascade. Located in apical plasma membrane and lateral plasma membrane. Is active in plasma membrane. Is expressed in several structures, including alimentary system; genitourinary system; heart; nervous system; and sensory organ epithelium. Human ortholog(s) of this gene implicated in lethal congenital contracture syndrome 2; lung adenocarcinoma; lung non-small cell carcinoma; and neuronal intestinal dysplasia type A. Orthologous to human ERBB3 (erb-b2 receptor tyrosine kinase 3).
