Assay PrincipleThis kit utilizes the double antibody sandwich enzyme-linked immunosorbent assay (ELISA) detection technique.Specific anti-human PCSK9 antibodies are precoated on a high-affinity ELISA plate. Standard samples, test samples, and biotinylated detection antibodies are added to the wells of the ELISA plate. After incubation, PCSK9 present in the samples binds to the solid-phase antibodies and the detection antibodies. After washing to remove unbound substances, streptavidin-HRP labeled with horseradish peroxidase is added. After washing, a colorimetric substrate, TMB, is added and the plate is incubated in the dark for color development. The intensity of the color reaction is directly proportional to the concentration of PCSK9 in the samples. A stop solution is added to terminate the reaction, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm).

Human PCSK9 Predicted Functions

Enables several functions, including lipoprotein particle receptor binding activity; serine-type endopeptidase activity; and sodium channel inhibitor activity. Involved in several processes, including cholesterol homeostasis; low-density lipoprotein particle receptor catabolic process; and regulation of receptor-mediated endocytosis. Located in several cellular components, including Golgi apparatus; endosome; and perinuclear region of cytoplasm. Part of PCSK9-AnxA2 complex and PCSK9-LDLR complex. Implicated in familial hypercholesterolemia and hypobetalipoproteinemia.

Mouse Pcsk9 Predicted Functions

Enables apolipoprotein binding activity; lipoprotein particle binding activity; and low-density lipoprotein particle receptor binding activity. Involved in several processes, including cellular response to insulin stimulus; cellular response to starvation; and regulation of neuron apoptotic process. Acts upstream of or within several processes, including low-density lipoprotein receptor particle metabolic process; regulation of low-density lipoprotein particle receptor catabolic process; and triglyceride metabolic process. Located in COPII-coated ER to Golgi transport vesicle; endoplasmic reticulum; and extracellular space. Is expressed in several structures, including alimentary system; cerebellum; genitourinary system; liver; and telencephalon. Human ortholog(s) of this gene implicated in familial hypercholesterolemia and hypobetalipoproteinemia. Orthologous to human PCSK9 (proprotein convertase subtilisin/kexin type 9).
