Assay Principle | This kit utilizes the double antibody sandwich enzyme-linked immunosorbent assay (ELISA) detection technique.Specific anti-mouse IL-12p70 antibodies are precoated on a high-affinity ELISA plate.Standards and test samples are added to the wells of the ELISA plate. After incubation, the IL-12p70 present in the samples binds to the solid-phase antibodies. After washing to remove unbound substances, biotinylated detection antibodies are added and incubated. After washing to remove unbound biotinylated antibodies, streptavidin-HRP labeled with horseradish peroxidase is added. After washing again, a signal enhancer is added and incubated. After washing to remove unbound substances, Streptavidin-HRP is added once more. After washing, a colorimetric substrate, TMB, is added and the plate is incubated in the dark for color development. The intensity of the color reaction is directly proportional to the concentration of IL-12p70 in the samples.A stop solution is added to terminate the reaction, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm). |

Human IL12B Predicted Functions
Enables identical protein binding activity; interleukin-12 alpha subunit binding activity; and protein heterodimerization activity. Contributes to cytokine activity; growth factor activity; and interleukin-23 receptor binding activity. Involved in several processes, including positive regulation of lymphocyte proliferation; positive regulation of peptidyl-tyrosine phosphorylation; and regulation of cytokine production. Acts upstream of or within negative regulation of protein secretion. Part of interleukin-12 complex and interleukin-23 complex. Implicated in asthma; chronic obstructive pulmonary disease; immunodeficiency 29; and salmonellosis. Biomarker of anogenital venereal wart.

Mouse Il12b Predicted Functions
Enables identical protein binding activity and interleukin-12 alpha subunit binding activity. Contributes to cytokine activity and cytokine receptor binding activity. Involved in positive regulation of T cell mediated cytotoxicity; positive regulation of T-helper 1 type immune response; and positive regulation of defense response to virus by host. Acts upstream of or within several processes, including cellular response to lipopolysaccharide; defense response to other organism; and positive regulation of interferon-gamma production. Located in cell surface; cytoplasm; and extracellular space. Part of interleukin-12 complex and interleukin-23 complex. Is expressed in colon. Human ortholog(s) of this gene implicated in asthma; chronic obstructive pulmonary disease; immunodeficiency 29; and salmonellosis. Orthologous to human IL12B (interleukin 12B).
