Assay PrincipleThis kit utilizes a solid-phase competitive enzyme-linked immunosorbent assay (ELISA) technique. Rabbit anti-mouse antibodies are precoated on a high-affinity ELISA plate. Specific monoclonal antibodies against PGE2 are added to the wells and incubated to bind to the immobilized antibodies. After washing, biotinylated PGE2 and unlabeled PGE2 or samples are added, competing for limited binding sites on the monoclonal antibodies. After washing to remove unbound substances, HRP-labeled streptavidin is added. After washing again, a chromogenic substrate is added for color development, and the intensity of the color reaction is inversely proportional to the concentration of PGE2 in the sample. The reaction is terminated by adding a stop solution, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm). This assay kit has been validated for human, mouse, and rat serum samples, but it is expected to detect PGE2 from other mammalian species as well.
