Assay Principle | This kit utilizes a competitive enzyme-linked immunosorbent assay (ELISA) technique. Rabbit anti-mouse antibody is precoated on a high-affinity ELISA plate. Cortisol-specific monoclonal antibodies are added to the wells of the plate and incubated, allowing them to bind to the solid-phase antibodies. After washing, HRP-labeled cortisol and unlabeled cortisol or samples are added, competing for limited binding sites on the monoclonal antibody. After washing to remove unbound substances, a chromogenic substrate is added, and the intensity of the color reaction is inversely proportional to the concentration of cortisol in the sample. The reaction is terminated by adding a stop solution, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm). This assay kit has been validated for human, mouse, and rat serum samples, but cortisol from other species is also expected to be detectable. |
