Assay PrincipleThis kit utilizes a solid-phase competitive enzyme-linked immunosorbent assay (ELISA) technique. Specific monoclonal antibodies against human ET-1 are precoated on a high-affinity ELISA plate. Biotinylated ET-1 and unlabeled ET-1 or samples are added into the wells of the ELISA plate, competing to bind to the limited binding sites on the coated antibody. After washing to remove unbound substances, HRP-labeled streptavidin is added. After washing again, TMB chromogenic substrate is added for color development while avoiding light. The intensity of the color reaction is inversely proportional to the concentration of ET-1 in the sample. The reaction is terminated by adding a stop solution, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm).

Human EDN1 Target Information

Human EDN1 Predicted Functions

Enables signaling receptor binding activity. Involved in several processes, including intracellular signal transduction; regulation of macromolecule biosynthetic process; and vascular associated smooth muscle contraction. Located in cytoplasm and extracellular space. Implicated in several diseases, including artery disease (multiple); bronchial disease (multiple); obesity; obstructive sleep apnea; and rheumatoid arthritis. Biomarker of several diseases, including artery disease (multiple); autoimmune disease (multiple); glaucoma (multiple); lung disease (multiple); and retinal disease (multiple).

Mouse Edn1 Target Information

Mouse Edn1 Predicted Functions

Enables endothelin A receptor binding activity. Involved in several processes, including cellular response to gonadotropin stimulus; circulatory system development; and neuron differentiation. Acts upstream of or within several processes, including G protein-coupled receptor signaling pathway; animal organ development; and rhythmic excitation. Located in extracellular space. Is expressed in several structures, including branchial arch; cardiovascular system; central nervous system; gut; and sensory organ. Human ortholog(s) of this gene implicated in several diseases, including artery disease (multiple); lung disease (multiple); obesity; obstructive sleep apnea; and rheumatoid arthritis. Orthologous to human EDN1 (endothelin 1).
