Assay PrincipleThis kit utilizes the double antibody sandwich enzyme-linked immunosorbent assay (ELISA) detection technique.Specific anti-human MMP-9 antibodies are precoated on a high-affinity ELISA plate. Standard samples, test samples, and biotinylated detection antibodies are added to the wells of the ELISA plate. After incubation, MMP-9 present in the samples binds to the solid-phase antibodies and the detection antibodies. After washing to remove unbound substances, streptavidin-HRP labeled with horseradish peroxidase is added. After washing, a colorimetric substrate, TMB, is added and the plate is incubated in the dark for color development. The intensity of the color reaction is directly proportional to the concentration of MMP-9 in the samples.A stop solution is added to terminate the reaction, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm).

Human MMP9 Predicted Functions

Enables identical protein binding activity; metalloendopeptidase activity; and serine-type endopeptidase activity. Involved in several processes, including cellular response to UV-A; negative regulation of apoptotic signaling pathway; and positive regulation of keratinocyte migration. Located in extracellular space. Implicated in several diseases, including Down syndrome; anodontia; artery disease (multiple); autoimmune disease (multiple); and chronic obstructive pulmonary disease (multiple). Biomarker of several diseases, including artery disease (multiple); autoimmune disease (multiple); brain disease (multiple); carcinoma (multiple); and lung disease (multiple).

Mouse Mmp9 Target Information

Mouse Mmp9 Predicted Functions

Enables metalloendopeptidase activity. Involved in negative regulation of epithelial cell differentiation involved in kidney development and response to amyloid-beta. Acts upstream of or within several processes, including collagen catabolic process; embryo implantation; and extracellular matrix organization. Located in collagen-containing extracellular matrix. Is expressed in several structures, including alimentary system; central nervous system; early conceptus; limb mesenchyme; and skeleton. Human ortholog(s) of this gene implicated in several diseases, including Down syndrome; anodontia; artery disease (multiple); autoimmune disease (multiple); and lung disease (multiple). Orthologous to human MMP9 (matrix metallopeptidase 9).

Rat Mmp9 Target Information

Rat Mmp9 Predicted Functions

Enables fibronectin binding activity and metallopeptidase activity. Involved in several processes, including cellular response to cell-matrix adhesion; cellular response to cytokine stimulus; and cellular response to iron(III) ion. Located in extracellular space. Part of protein-containing complex. Used to study artery disease (multiple); brain ischemia (multiple); congestive heart failure; glomerulosclerosis; and lung disease (multiple). Biomarker of several diseases, including artery disease (multiple); brain disease (multiple); chronic obstructive pulmonary disease (multiple); glucose metabolism disease (multiple); and kidney disease (multiple). Human ortholog(s) of this gene implicated in several diseases, including Down syndrome; anodontia; artery disease (multiple); autoimmune disease (multiple); and chronic obstructive pulmonary disease (multiple). Orthologous to human MMP9 (matrix metallopeptidase 9).
