Assay PrincipleThis kit utilizes the double antibody sandwich enzyme-linked immunosorbent assay (ELISA) detection technique.Specific anti-human IGFBP-3 antibodies are precoated on a high-affinity ELISA plate. Standard samples, test samples, and biotinylated detection antibodies are added to the wells of the ELISA plate. After incubation, IGFBP-3 present in the samples binds to the solid-phase antibodies and the detection antibodies. After washing to remove unbound substances, streptavidin-HRP labeled with horseradish peroxidase is added. After washing, a colorimetric substrate, TMB, is added and the plate is incubated in the dark for color development. The intensity of the color reaction is directly proportional to the concentration of IGFBP-3 in the samples.A stop solution is added to terminate the reaction, and the absorbance value is measured at a wavelength of 450 nm (with a reference wavelength range of 570-630 nm).

Human IGFBP3 Predicted Functions

Enables insulin-like growth factor I binding activity and protein tyrosine phosphatase activator activity. Involved in several processes, including negative regulation of smooth muscle cell migration; negative regulation of smooth muscle cell proliferation; and positive regulation of myoblast differentiation. Acts upstream of or within negative regulation of cell population proliferation; negative regulation of protein phosphorylation; and protein phosphorylation. Located in extracellular space and nucleus. Part of insulin-like growth factor ternary complex. Implicated in several diseases, including Alzheimer’s disease; Turner syndrome; diabetes mellitus (multiple); prostate cancer; and transitional cell carcinoma. Biomarker of several diseases, including Barrett’s esophagus; Down syndrome; autoimmune disease (multiple); bone disease (multiple); and urinary system cancer (multiple).

Mouse Igfbp3 Target Information

Mouse Igfbp3 Predicted Functions

Enables fibronectin binding activity and insulin-like growth factor binding activity. Acts upstream of or within several processes, including osteoblast differentiation; positive regulation of signal transduction; and type B pancreatic cell proliferation. Located in extracellular space. Is expressed in several structures, including alimentary system; brain; early embryo; gonad; and limb. Human ortholog(s) of this gene implicated in several diseases, including Alzheimer’s disease; Turner syndrome; diabetes mellitus (multiple); prostate cancer; and transitional cell carcinoma. Orthologous to human IGFBP3 (insulin like growth factor binding protein 3).

Rat Igfbp3 Target Information

Rat Igfbp3 Predicted Functions

Enables insulin-like growth factor II binding activity. Involved in several processes, including cellular response to Thyroglobulin triiodothyronine; cellular response to estradiol stimulus; and response to peptide hormone. Located in extracellular space; heterochromatin; and platelet alpha granule. Part of insulin-like growth factor binary complex and insulin-like growth factor ternary complex. Used to study impotence. Biomarker of congenital diaphragmatic hernia; impotence; and obesity. Human ortholog(s) of this gene implicated in several diseases, including Alzheimer’s disease; Turner syndrome; diabetes mellitus (multiple); prostate cancer; and transitional cell carcinoma. Orthologous to human IGFBP3 (insulin like growth factor binding protein 3).
